TAE buffer is a common electrophoretic buffer composed of Tris base, acetic acid, and EDTA. It is often used as a buffer for gel electrophoresis of DNA or RNA in molecular biology experiments.
TAE buffer has the following characteristics:
Provide conductivity and maintain pH value: Tris acetate can maintain the pH value of the solution while providing conductivity.
Inhibition of metal dependent nucleases: EDTA in TAE buffer inhibits metal dependent nucleases by chelating divalent cations (Ca2+, Mg2+), thereby protecting DNA from the influence of nucleases during operation.
It is applicable to the application of DNA fragments that need to be modified by DNA modifying enzymes for gel elution: in this case, the use of TBE buffer should be avoided, because the borate of TBE buffer will inhibit many enzymes (such as DNA ligase).
However, the buffering capacity of TAE buffer is lower than that of TBE, so it should be avoided for long-term and repetitive electrophoresis.
