3-(4-Chlorbutyl)-1H-indol-5-carbonitril is a key intermediate for synthetizing Vilazodone.
Vilazodone is developed by Clinical Data. Originally, its chemical compound was developed by the German company Merck KGaA. Vilazodone is a serotonergic antidepressant for treating major depressive disorder, and was approved by the FDA for use in the United States in 2011. It is produced by Trovis Pharmaceuticals LLC under the trade name Viibryd. There are 3 specifications of 10mg, 20mg, and 40mg with the Vilazodone tablet approved in the market.
Vilazodone hydrochloride is synthesized from 5-(1-Piperazinyl)benzofuran-2-carboxamide and 3-(4-Chlorobutyl)-1H-indole-5-carbonitrile by condensation and salification.

Preparation of 3-(4-Chlorbutyl)-1H-indol-5-carbonitril
3-(4-Chlorbutyl)-1H-indole-5-carbonitrile is prepared from 5-cyanoindole and 4-chlorobutyl chloride by acylation and reduction.
